let me make this a little clearer maybe....it was my fuel injection pump that the fuel screw is located on. i took the cap off to reveal that spring loaded membrane. when i put the cap back on it wouldnt start. ...could i have rotated it by accident which made it not start?

Check to make sure both wires are hooked up to the fuel shut-off solenoid behind and just below the power screw on the backside of the injection pump.

I agree, fuel selinoid on the back of the IP. Must have 12V to start. Are you refering to the very top cap that has 3 allens and one flat, or 4 flats if it's non IC or manual? The spring loaded membrane you're reffering to is the diaphram, attached is the fuel pin and underneath is the starwheel. Tampering with these will not cause a NO Start.

Did you check the wires on the back side of the IP?

There is no issue with removing the fuel pin and diaphram and reinstalling. The only thing that could cause a no start would be fuel supply. It has to have 12V dc at the terminals below the AFC air line that is attached to the cylinder head from the pump top that you removed.
